dc.description.abstractPara avaliar os aspectos parasitológicos, epidemiológicos, clínicos, laboratoriais e anátomo-histopatológicos da doença de Dalmeny, numa primeira etapa caninos com 6 meses de idade foram infectados com musculatura estriada cardíaca bovina contendo sarcocistos, para obtenção de esporocistos de Sarcocystis. Nos cães infectados não foram observadas alterações clínicas ou patológicas, assinalando-se, porém, a presença de esporocistos nos linfonodos mesentéricos. Com os esporocistos colhidos das fezes dos cães, numa segunda etapa bezerros com 3 a 6 meses de idade foram infectados com inóculos contendo 250 mil, 500 mil ou 1 milhão de esporocistos, e sinais clínicos relevantes foram observados no período de 24 a 28 dias após a infecção (DPI), independente do inóculo administrado. Merontes de 1ª e 2ª gerações foram encontrados em animais que morreram entre o 13º e o 30º DPI e merontes e merozoítas de 3ª geração foram encontrados em cé- lulas mononucleares, em esfregaços de sangue periférico ou da cepa de leucócitos, preparados do 24º ao 27º DPI. Com ba- XXiii se no hemograma completo caracterizou-se, na série vermelha, uma anemia macrocítica com resposta, e na série branca, leucocitose por aumento nos números de linfócitos e neutrófilos. Níveis de proteína plasmática total diminuíram, enquanto de fibrinogênio aumentaram, progressivamente, da 1ª à 8ª semana após a infecção (SPI). Atividades de ASAT e CPK tiveram aumentos do 27º ao 48º DPI, enquanto em LDH ocorreram do 13º ao 41º DPI. Achados anátomo-patológicos mais importantes foram caracterizados como linfadenopatia generalizada, palidez de mucosas, emaciação, atrofia serosa da gordura, excessivo volume líquido amarelo nas cavidades pericárdica, pleural e peritoneal, sangue ralo, petéquias e sufusões em serosas de muitos órgãos, e musculatura estriada esquelética de aspecto tigróide. Histopatologicamente observaram-se esquizontes e infiltrados celulares mononucleares em vários órgãos examinados de 24 a 48 DPI, hemorragias no sistema nervoso central, nefrite não supurativa, atrofia de glomérulo, e cistos maduros, redondos ou fusiformes, de parede lisa e medindo menos de 0.5 micra de espessura, característicos da espécie S. cruzi. Bradizoítas foram obtidos após digestão tríptica de músculo cardíaco de animal morto aos 100 DPI, sendo a técnica segura, com a vantagem de dar resultados de forma mais rápida do que a digestão pépticapor

dc.description.abstractOtherTo evaluante the parasitological, epidemiological and pathological aspects of Dalmeny's disease, 15 dogs with 6 months old approximately were fed with bovine cardiac muscle, positive for sarcocystis in order to obtain sporocysts. After 11 to 17 days, sporocysts of Sarcocystis cruzi with 15x10µm in diameters were shed by the dogs. The prepatent period was 11 to 17 days while the patent period was 27 days. However, when some of the dogs were infected again with positive cardiac muscle, the prepatent and patent periodes were 7 to 12 and 60 days, respectively. No Clinical sings nor pathologic findings were observed, except the appearance of sporocysts, well developed, in the mesenteric linph nodes of 4 dogs necropsyed in the end of the experiment. With the sporocysts obtained at the Ist part of the experiment, 3 to 6 months calves were infected with 25x104, 5x105 or 106 sporocysts and significant clinical signs ocur- XXV red without regard to the inoculum, and the parasitological findings were characterized by the appearance of meronts in the endotelial cells of the cappilaries in different organs, and merozoites of 3rd generation in mononuclear cells of blood smears or in buffy coat from 24 to 27 days after infection (DAI). The erythrocitic values were characterized by macrocytic anemia while the leucocytic values were consisted by leucocytosis with an increase of lymphocytes and neutrophils. When levels of total plasmatic protein decreased, fibrinogen increased progressively from 1 st to 8 th week post infection (WPI). Increased activities of ASAT and CPK were observed from 27 th to 48 th DAI while the values of LDH had increased from 13 th to 41 th DAI. The clinical signs were prominent from 24 to 28 DAI and were consisted by anorexia, lost of weight, alopecia, dyspnea, lacrimation, salivation, yellowish discharge from the nostrils, prostration and hypothermia before death, while the necropsy findings were characterized by pale mucous membranes, serous atrophy of fat, excessive yellowish fluid in the pericardial, thoraxic and peritoneal cavities, and aqueous blood. Petequeas and sufusions were observed above the serosa of different organs. Further, the striated muscle has a striped appearance. Microscopically, lesions were consisted of hemorrhage around blood vessels in different organs. The whiteness of muscle was characterized by necrosis with hyalini- xxvi zation or fragmentation of the muscle fibers. The kidneys had diffuse lesions such as subcapsular hemorrhage around blood vessels, and congestion of cortical and medular capillaries. At this stage the renal interstitium contained mononuclear cells infiltration. Casts of amorphous material were founded in the renal tubules, and glomerular capsules were thickened by increasing protein-like material deposited on the external surface of the basement membrane.eng
