dc.description.abstractOs ectoparasitos de cães e gatos são pulgas, carrapatos, moscas, piolhos e sarnas. As infestações decorrentes destes parasitos podem levar a infecções graves tanto para os animais como para seres humanos. O objetivo deste trabalho foi avaliar as contribuições do sarolaner nos tratamentos clínicos de infestações controladas de Ctenocephalides felis felis em cães e gatos, Rhipicephalus sanguineus em cães e no tratamento de miíases de ocorrência natural causadas por Cochliomyia hominivorax em cães. Sendo assim, quatro estudos foram realizados e separados em capítulos. Os capítulos foram intitulados I “Eficácia do sarolaner no controle de carrapatos Rhipicephalus sanguineus em cães infestados artificialmente”, II “Eficácia do sarolaner no controle de pulgas Ctenocephalides felis felis em cães infestados artificialmente”, III “Eficácia do sarolaner no controle de pulgas Ctenocephalides felis felis em gatos infestados artificialmente” e IV “Eficácia do sarolaner no tratamento de miíases causadas por Cochliomyia hominivorax em cães naturalmente infestados”. No estudo de eficácia carrapaticida foram selecionados 20 cães beagles adultos. As infestações foram realizadas com 25 machos e 25 fêmeas adultos não alimentados de R. sanguineus semanalmente por 5 semanas. As contagens foram realizadas semanalmente 48 horas após as infestações. Os animais receberam 2,0 a 4,0mg/kg do sarolaner por via oral em dose única. A eficácia média do sarolaner foi 97,3%, por 35 dias. Para eficácia pulguicida em cães foram incluídos 20 animais da raça Beagle, machos e fêmeas, com idade entre 1 e 4 anos que receberam por via oral 2,0 a 4,0mg/kg do sarolaner em dose única. As infestações ocorreram semanalmente com 50 (+-5) casais de pulgas C felis adultas não alimentadas. As desinfestações ou penteações foram realizadas 24 horas após as infestações. A média de eficácia foi de 99,75% no período de 35 dias. Para avaliação da eficácia pulguicida em gatos, foram incluídos 14 animais sem raça definida machos e fêmeas em igual proporção, idade entre um e seis anos. Os animais receberam 2,0-3,5mg/kg de sarolaner por via oral em dose única. As infestações foram realizadas semanalmente com 50(+-5) casais de pulgas C felis por 12 semanas. As contagens ocorreram 48 horas após as infestações e a eficácia do sarolaner foi de 100% em todos os dias experimentais, totalizando 84 dias. No tratamento de miíases de ocorrência natural por Cochliomyia hominivorax foram avaliados oito cães beagles adultos que receberam a dose de 2,0 – 4,0mg/kg de sarolaner por via oral e a medicação de suporte para tratamento da ferida. A expulsão das larvas foi avaliada a cada 15 minutos na primeira hora e a cada hora nas seis horas subsequentes e com 24 horas de tratamento. As larvas foram avaliadas quanto à motilidade para diferenciação entre mortas e vivas. Após 24 horas os animais foram sedados para a limpeza da ferida e a retirada manual de todas as larvas. A média de expulsão das larvas foi de 50,9% ocorrendo principalmente após quatro horas do tratamento. O efeito larvicida foi de 70,6% nas seis horas após o tratamento e de 100% em 24 horas. A eficácia global foi de 100%. O sarolaner se mostrou eficaz em todos os tratamentos propostos podendo ser uma boa escolha em casos de infestações múltiplas. Não foram observadas alterações clinicas ou comportamentais durante os estudos que pudessem estar relacionadas ao tratamento ou aos procedimentos realizadospor

dc.description.abstractOtherDog and cat ectoparasites are fleas, ticks, flies, lice and scabies. Infestation from these parasites can lead to serious infections for both animals and humans. The aim of this study was to evaluate the contributions of sarolaner in the clinical treatment of controlled Ctenocephalides felis felis infestations in dogs and cats, Rhipicephalus sanguineus in dogs and in the treatment of naturally occurring myiasis caused by Cochliomyia hominivorax in dogs. Thus, four studies were conducted and separated into chapters. The chapters were titled I “Efficacy of sarolaner in the control of Rhipicephalus sanguineus ticks in artificially infested dogs”, II “Efficacy of sarolaner in the control of Ctenocephalides felis felis fleas”, III “Efficacy of sarolaner in the control of Ctenocephalides felis fleas felis in artificially infested cats ”and IV“ Effectiveness of sarolaner in the treatment of myiasis caused by Cochliomyia hominivorax in naturally infested dogs ”. In the tick efficacy study, 20 adult beagles were selected. Infestations were carried out with 25 male and 25 non-fed R. sanguineus adult females weekly for 5 weeks. Counts were performed weekly 48 hours after infestations. The animals received 2.0 to 4.0 mg/kg of sarolaner orally in a single dose. The average efficacy of sarolaner was 97.3% for 35 days. For efficacy in dogs, 20 male and female Beagle animals, aged between 1 and 4 years, who received orally 2.0 to 4.0 mg/kg of sarolaner in a single dose. Infestations occurred weekly with 50 (+ -5) pairs of unfeeded adult C felis. Desinfestations or combing were performed 24 hours after the infestations. The average efficacy was 99.75% over the 35 day period. To evaluate the efficacy of cats, 14 male and female mongrel animals were included in the same proportion, aged between one and six years. The animals received 2.0 to 3.5 mg/kg sarolaner orally in a single dose. Infestations were performed weekly with 50 (+/- 5) C flea couples for 12 weeks. Counts occurred 48 hours after infestations and sarolaner was 100% effective on all experimental days, totaling 84 days. In the treatment of naturally occurring myiasis by Cochliomyia hominivorax, eight adult beagle dogs receiving oral 2.0 to 4.0mg/kg sarolaner and supportive wound care medication were evaluated. Larval expulsion was evaluated every 15 minutes in the first hour and every hour for the next six hours and with 24 hours of treatment. Larvae were evaluated for motility for differentiation between dead and alive. After 24 hours the animals were sedated for wound cleaning and manual removal of all larvae. The average expulsion of the larvae was 50.9% occurring mainly after four hours of treatment. The larvicidal effect was 70.6% at six hours after treatment and 100% at 24 hours. Overall efficacy was 100%. Sarolaner was effective in all proposed treatments and may be a good choice in cases of multiple infestations. No clinical or behavioral changes were observed during studies that could be related to treatment or procedures performedeng
