dc.description.abstractO presente trabalho teve como objetivo diagnosticar as larvas de nematoides anisaquídeos em diferentes hospedeiros no litoral do Estado do Rio de Janeiro, Brasil. Entre março de 2013 e setembro de 2014 foram feitas necropsias de 146 espécimes de peixes, pertencentes à cinco ordens e dez famílias na costa de Cabo Frio (23°S, 42°W), Rio de Janeiro. No capítulo 1, as três espécies de Pinguipedidae (Actinopterygii, Perciformes) que ocorrem no Brasil, Pseudopercis numida Miranda-Ribeiro, 1903, P. semifasciata (Cuvier, 1829) e Pinguipes brasilianus Cuvier, 1829, foram examinadas para coleta de larvas de anisaquídeos. Nenhum anisaquídeo foi encontrado em P. semifasciata. Larvas de Anisakis typica (Diesing, 1860) e Hysterothylacium sp. foram reportadas em P. numida. Larvas de Raphidascaris sp. e Hysterothylacium sp. foram reportadas em P. brasilianus. Pseudopercis numida é um novo registro de hospedeiro para A. typica. Não foi possível identificar as espécies de Hystertothylacium e Raphidascaris devido à ausência de sequências similares de adultos depositadas no GenBank. No capítulo 2, larvas de Hysterothylacium spp. coletadas em Caulolatilus chrysops (Valenciennes, 1833), Gymnothorax vicinus (Castelnau, 1855), Mentichirrus americanus (Linnaeus, 1758), Merluccius hubbsi Marini, 1933, Mullus argentinae Hubbs & Marini, 1933, P. brasilianus, Priacanthus arenatus Cuvier, 1829, Prionotus punctatus (Bloch, 1793), Sarda sarda (Bloch, 1793), e Zenopsis conchifer (Lowe, 1852) provenientes da mesma localidade foram caracterizadas geneticamente. Quatro morfotipos foram encontrados para três genótipos diferentes. Não foi possível identificar em nível de espécie nenhum morfotipo devido à ausência de sequências similares de espécimes adultos depositadas no GenBank. Foram obtidas sequências parciais cox-2 mtDNA e ITS 1, 5.8S e ITS 2 rDNA destas larvas e comparadas com outras sequências disponíveis no banco de dados do GenBank. Em ambos os estudos a morfologia foi avaliada juntamente com a genética. Nas análises de cox-2 mtDNA das larvas de Hysterothylacium sp. em P. numida (capítulo 1) e dos morfotipos 1 e 2 (capítulo 2) foram observadas alta similaridade entre nossas sequências de Hysterothylacium sp. com sequências de Hysterothylacium deardorffoverstreetorum Knoff, Felizardo, Iñiguez, Maldonado Jr, Torres, Pinto & Gomes, 2012. No entanto a análise filogenética mostrou a presença de clados distintos entre sequências de mesmo nome.por

dc.description.abstractOtherThe current work aimed to diagnose nematode anisakid larvae in different hosts from the coast of State of Rio de Janeiro, Brazil. Between March 2013 and September 2014 were made autopsies of 146 specimens of fish, belonging to five orders and ten families in Cabo Frio coast (23 ° S, 42 ° W) Rio de Janeiro State. In Chapter 1, the three species of Pinguipedidae (Actinopterygii, Perciformes) that occur in Brazil, Pseudopercis numida Miranda-Ribeiro, 1903, P. semifasciata (Cuvier, 1829) e Pinguipes brasilianus Cuvier, 1829, were examined for collection of anisakid larvae. Anisakid larvae were not found in P. semifasciata. Larvae of Anisakis typica (Diesing, 1860) and Hysterothylacium sp. were reported in P. numida. Larvae Raphidascaris sp. and Hysterothylacium sp. were reported in P. brasilianus. Pseudopercis numida is a new host record for A. typica. It was not possible to identify the species of Hystertothylacium Raphidascaris due to the lack of similar sequences of adults from GenBank database. In Chapter 2, larvae of Hysterothylacium spp. were collected in Caulolatilus chrysops (Valenciennes, 1833), Gymnothorax vicinus (Castelnau, 1855), Mentichirrus americanus (Linnaeus, 1758), Merluccius hubbsi Marini, 1933, Mullus argentinae Hubbs & Marini, 1933, P. brasilianus, Priacanthus arenatus Cuvier, 1829, Prionotus punctatus (Bloch, 1793), Sarda sarda (Bloch, 1793) and Zenopsis conchifer (Lowe, 1852) from the same locality were genetically characterized. In both studies the morphology was evaluated together with the genetic. In the analysis of cox-2 mtDNA larvae of Hysterothylacium sp. in P. numida (Chapter 1) and morphotypes 1 and 2 (Chapter 2) were observed high similarity between our sequences with sequences of Hysterothylacium deardorffoverstreetorum Knoff, Felizardo Iñiguez, Maldonado, Jr. Torres, Pinto & Gomes, 2012. However phylogenetic analysis showed the presence of distinct clades of sequences of the same name.eng
