| dc.description.abstract | As células dendríticas plasmocitoides (pDCs) constituem um subgrupo de células dendríticas precursoras presentes no sangue periférico e em órgãos linfoides. Essas células são identificadas pela alta expressão do receptor da cadeia α da Interleucina 3 (IL-3Rα) e produzem grandes quantidades de Interferon tipo I (IFN-I), mediando a resposta imune. Na imuno-histoquímica, o marcador utilizado para a detecção desse tipo celular é o anticorpo anti-CD123. As pDCs tornaram-se alvo de estudos na medicina humana para o diagnóstico e o desenvolvimento de terapias antitumorais de neoplasias de origem hematológica, processos inflamatórios e doenças autoimunes; pouco se sabe na medicina veterinária sobre a existência das células dendríticas plasmocitoides e sua distribuição em tecido linfoide normal. Em cães, há apenas um trabalho que empregou o anticorpo anti-CD123 para imunomarcação de pDCs em tumores mamários de cadelas, associando a sua presença com um pior prognóstico tumoral para essa espécie. Dessa forma, o estudo teve como objetivo verificar a eficiência do marcador para a detecção de células CD123+ em linfonodos de cães, gatos, bovinos e equinos e se, nessas espécies, a distribuição é similar à observada em humanos. Realizou-se exame imuno-histoquímico com anticorpo anti- CD123 monoclonal e policlonal em 10 amostras de linfonodos das espécies bovina, felina e canina, e em 5 amostras da espécie equina. Os resultados obtidos com o anticorpo monoclonal não foram satisfatórios. Já com o anticorpo policlonal, células CD123+ foram evidenciadas agrupadas ou individualizadas de forma predominante no paracórtex de todas as amostras de caninos (10/10) e em aproximadamente todos os linfonodos de bovinos (8/10). Na espécie felina os resultados foram negativos em todas as amostras, e em equinos os resultados foram desconsiderados pela presença acentuada de hemossiderina. Concluiu-se que o CD123 é um marcador válido para uso em caninos e bovinos e que estas células, assim como em humanos, distribuem-se mais frequentemente na região paracortical do linfonodo | pt_BR |

| dc.description.abstractOther | Plasmacytoid dendritic cells (pDCs) constitute a subgroup of precursor dendritic cells found in peripheral blood and lymphoid organs. These cells are characterized by the high expression of the Interleukin-3 α-chain receptor (IL-3Rα) and their capacity to produce large amounts of type- I Interferon (IFN-I), thereby orchestrating the immune response. The anti-CD123 antibody is the standard marker used for pDC detection in immunohistochemistry. While pDCs are a major focus in human medicine for the diagnosis and development of antitumor therapies against hematological malignancies, inflammatory processes, and autoimmune diseases, their existence and distribution in normal lymphoid tissue remain poorly understood in veterinary medicine. In dogs, only one study has utilized anti-CD123 antibodies for the immunolabeling of pDCs in mammary tumors, linking their presence to a worse prognosis. This study aimed to evaluate the efficiency of the anti-CD123 marker for detecting CD123+ cells in the lymph nodes of dogs, cats, cattle, and horses, and to determine if their distribution in these species is comparable to that observed in humans. Immunohistochemical analysis was performed using monoclonal and polyclonal anti-CD123 antibodies on 10 lymph node samples from bovine, feline, and canine species, and 5 samples from the equine species. The results obtained with the monoclonal antibodies were unsatisfactory. However, using polyclonal antibodies, CD123+ cells were predominantly observed, either grouped or individually, in the paracortex of all canine samples (10/10) and in most bovine lymph nodes (8/10). Results were negative in all cat samples. Equine samples were excluded from the final evaluation due to the marked presence of hemosiderin. In conclusion, CD123 is a valid immunohistochemical marker for pDCs in canines and bovines.
