dc.description.abstractOs probióticos são microrganismos vivos que, quando administrados em quantidades adequadas, conferem benefícios à saúde do hospedeiro. Tanto a nutrição quanto a microbiota materna podem modular a saúde fetal. No entanto, não se sabe os impactos da suplementação com probióticos no comportamento emocional da prole. Assim, o objetivo do estudo foi avaliar se a suplementação materna com bactérias lácticas é capaz de alterar parâmetros neurocomportamentais em prole de camundongos suíços. Para este propósito, camundongos prenhas (n = 6 cada, e ~ 35g) foram divididos aleatoriamente em dois grupos: controle e grupo tratado com probiótico. Estes animais foram tratados durante a gravidez e lactação com leite ou leite fermentado com Lactobacillus paracasei a uma concentração de 107 células / ml (p.o, diariamente). Após o término do tratamento, parte da progênie com 24 dias de vida foi submetida a eutanásia e o hipocampo foi dissecado para análise de RNA. Após completar 70 dias de vida, os demais filhotes foram submetidos a uma bateria de testes comportamentais composta pelos seguintes protocolos: campo aberto, caixa escura clara, testes de labirinto em cruz elevado e suspensão em cauda. De acordo com os resultados, os animais cujas mães foram tratadas com leite fermentado tiveram uma redução na expressão de GAD 65 (19%, p = 0,006), GAD 67 (25%, p = 0,003) e subunidade α3do receptor GABAA (223%, p = 0,01) no hipocampo. Na fase adulta, a prole do grupo tratado apresentou maior tempo na área central no teste de campo aberto (62%, p = 0,04), menor latência para o lado claro no teste de caixa claro-escuro (252% p = 0,01) e menor número de comportamentos de tomada de decisão no teste do labirinto em cruz elevado (30%, p = 0,02). Nenhum resultado significativo foi encontrado no teste de suspensão da cauda. Estes resultados indicam que a suplementação materna com probiótico modula o sistema gabaérgico hipocampal da prole. Tais mudanças podem estar associadas à redução do comportamento semelhante à ansiedade ao atingir a idade adulta.por

dc.description.abstractOtherProbiotics are living microorganisms which, when administered in suitable amounts, confer a health benefit on the host. Both nutrition and maternal microbiota can modulate fetal health. However, it is unknown whether probiotics could have a direct effect offspring emotional behavior. Thereby, the objective of our study was to evaluate whether maternal supplementation with acid lactic bacteria is able to alter neurobehavioral parameters of Swiss mice offspring. For this purpose, pregnant mice (n=6 each, and ~35g) were randomly divided into two groups: control and probiotic-treated group. These animals were treated during pregnancy and lactation with milk or milk fermented with Lactobacillus paracasei at a concentration of 107 cells/ml (p.o, daily). After the end of the treatment, part of the progeny with 21 days of life was submitted to euthanasia, and the hippocampus was dissected for RNA analysis. After completing 70 days of life, the rest of the pups were submitted to a battery of behavioral tests composed of the following protocols: open field, light dark box, elevated plus maze and tail suspension tests. According to our results, animals whose mothers were treated with fermented milk had a reduction in the expression of GAD 65 (19%, p = 0.006), GAD 67 (25%, p = 0.003) and α3 subunit GABAA (223%, p = 0.01) in the hippocampus. In the adult phase, the offspring of the treated group had higher time in the central area in the open field test (62%, p = 0.04), lower latency to the light side in the light-dark box test (252% p = 0.01) and lower number of decision-making behaviors in the elevated plus maze test (30%, p = 0.02). No significant results were found in the tail suspension test. These results indicate that maternal supplementation with probiotic modulates offspring hippocampal gabaergic system. Such changes could possibly be associated with the reduction of anxiety-like behavior upon reaching adulthood.eng
