Considering the importance of conservation of Cariniana legalis coupled with ample possibil-ities of use and marketing, the objective of this work was to perform the in vitro cultivation of this species and compare the anatomical structures leaves with plants grown in a vegetation house. For the evaluations were used leaves of seedlings of in vitro cultivation kept 15 days in house B.O.D. with temperature of 25ºC and absence of light, of seedlings transferred to growth room at 25 ± 2°C, photoperiod of 16 hours and irradiance of 35 μmol m-1 s-1, where they remained for 15 days, seedling emergence in good stage of development which were selected and transferred to the conditions of acclimatization using plastic bags are transparent and in a shade house with commercial substrate Mecplant®, where they remained for 21 days and leaves of plants cultivated ex vitro. The anatomical study was made from photomicro-graphs of transversal sections and paradermics leaves, examined in microscopic, and the measured adatial and abbatial epiderms tissue, palisade and sponges parenchyma, mesophly, largest in limb, central nervure, polar and equatorial diameter of stomata and sto-matal density through software AxionVision. The data were subjected to analysis of variance and means were compared by the Tukey test. The leaf structures of Cariniana legalis feature dorsiventral organization, uniseriate epidermis hipostomatic, and the stomate of type anomo-cytic. Leaves of cultivation in a greenhouse, in comparison to the cultures in vitro, showed greater thickness of the adatial epidermis and central nervure and presence of sclerenchyn. Sheets from the in vitro cultivation, increased thickness of the largest in limb, stomata larger and in greater numbers, but less functional that the stomata of the leaves of the ex vitro and plant cultivation climatized. The plants of Cariniana legalis demonstrated plasticity to adapt the variations of irradiance of light, humidity and temperature in the environmental conditions to which they were subjected, this fact of great importance for a good development ex vitro.pt_BR
